In May 2026, the annual meeting of the Association for Research in Vision and Ophthalmology (ARVO) concluded in Denver, USA.

At this premier academic event gathering cutting-edge ophthalmology research from around the world, Frontera Therapeutics delivered a "double-barreled" presentation, disclosing positive interim Phase II clinical data for its core products FT-002 and FT-003 in two oral presentations.

As the only company in China to have advanced an X-linked retinitis pigmentosa (XLRP) gene therapy into Phase II clinical trials, and a leader in the intravitreal gene therapy space for nAMD, Frontera is leveraging solid clinical data and proprietary technology platforms to accelerate its capture of a position in the global gene therapy competitive landscape.

TONACEA 01: FT-002 – The First Domestic XLRP Gene Therapy to Enter Phase II

FT-002 is a recombinant adeno-associated virus (rAAV) gene therapy candidate targeting XLRP.

XLRP is one of the most common and severe forms of inherited retinal disease. Patients typically develop symptoms before the age of 10 and progress to legal blindness by age 50. According to Frost & Sullivan data, in 2024, there were approximately 62,000 XLRP patients in China and 15,000 in the US. No approved effective therapy exists globally.

This field was once a battleground for multinational giants. In 2019, Biogen acquired Nightstar Therapeutics for US877 million, only to announce in 2021 that the Phase II/III trial did not meet its primary endpoint. In 2023, Johnson & Johnson licensed bota-vec from MeiraGTx for a US130 million upfront payment, but in 2025, its Phase III LUMEOS study also failed to meet its primary endpoint.

With over US$1 billion in combined investments faltering, XLRP has become known in the industry as a "high-difficulty examination hall" for AAV gene therapy.

Against this backdrop of setbacks, FT-002 stands as one of the few remaining assets in the field. It is the first and only XLRP rAAV gene therapy candidate to enter clinical development in China, having advanced to Phase II. Concurrently, FT-002 has also received a Phase II clinical trial approval in the US, having previously been granted Orphan Drug Designation and Fast Track Designation by the FDA.

At this year's ARVO meeting, Frontera presented positive updated Phase II results for FT-002 in pediatric XLRP subjects, with several core metrics showing notable improvements:

  • Low Luminance Visual Acuity (LLVA): 50% of subjects achieved a ≥10-letter improvement, and 33% achieved a ≥15-letter improvement.

  • Best-Corrected Visual Acuity (BCVA): Data from completed Phase I study showed an average BCVA improvement of 8.3±6.11 letters in the high-dose group. 33.3% of subjects achieved a ≥15-letter gain, with sustained BCVA improvement for over 52 weeks, suggesting FT-002 also has the potential to restore or preserve central vision.

  • Retinal Function Indicators: Overall showed a sustained trend of improvement.

  • Safety: No drug-related dose-limiting toxicities or serious adverse events were observed.

The improvement in LLVA has an immeasurable impact on the quality of life for XLRP patients. Early symptoms of the disease are often visual difficulties in low-light environments, with many children experiencing problems at dusk or during indoor activities. Gains in LLVA mean a potentially substantial improvement in patients' basic daily living abilities, such as nighttime walking and indoor mobility.

Looking at the XLRP gene therapy competitive landscape, FT-002 is not without competition. One of its competitors is laruzova, an AAV gene therapy from Beacon Therapeutics. Its Phase II study showed sustained visual benefit, with topline data from the Phase II/III VISTA trial expected in the second half of 2026.

TONACEA 02: FT-003 – Sustained Benefit from a Single Injection

If FT-002 targets a gap in the rare disease space, FT-003 confronts a much larger battlefield: neovascular age-related macular degeneration (nAMD) and diabetic macular edema (DME).

nAMD and DME are two leading causes of vision loss in the elderly worldwide, with patient populations growing as the population ages and diabetes incidence rises.

Current standard-of-care for nAMD and DME relies primarily on frequent intravitreal injections of anti-VEGF agents, typically every 4 to 8 weeks. This high-frequency dosing regimen imposes a heavy burden on patients, leads to poor compliance, and results in substantial healthcare system costs. According to a cost-effectiveness study, the total cost of long-term anti-VEGF therapy can reach up to US$304,000 in the US. There is a pressing market need for long-acting therapies offering "sustained control from a single dose."

FT-003 achieves long-term expression of aflibercept via a single intravitreal injection, aiming to fundamentally eliminate the need for frequent repeat injections. It is one of the most advanced intravitreal gene therapy programs globally for nAMD and DME, and the fastest-advancing candidate for intravitreal administration in China.

Data presented at ARVO further solidified and highlighted FT-003's differentiated positioning.

  • In nAMD patients, FT-003 reduced annual anti-VEGF injection frequency by over 90% compared to the control group.

  • In the two FT-003 dose groups, 85.0% and 78.9% of subjects, respectively, required no rescue injections over 28 weeks, demonstrating a significant long-term treatment burden reduction.

  • Throughout the 52-week follow-up period, FT-003 showed a favorable safety profile, with no infectious endophthalmitis, vasculitis, vascular occlusion, or FT-003-related serious adverse events observed.

Longer-term follow-up data is equally robust. Frontera data show that some patients who received FT-003 have not required any anti-VEGF treatment for up to three years, while maintaining stable and improved visual acuity and retinal structure.

Globally, the gene therapy race in nAMD/DME is heating up significantly.

  • ABBV-RGX-314, from AbbVie and REGENXBIO, is delivered via suprachoroidal injection and has advanced to Phase III clinical trials, targeting both wet AMD and diabetic retinopathy.

  • Another giant, Eli Lilly, acquired Adverum Biotechnologies in October 2025, adding Ixo-vec (an intravitreal gene therapy) to its pipeline. The Phase III ARTEMIS trial has completed screening, with topline readings expected in Q1 2027.

  • 4DMT's 4D-150 (dual transgene therapy) has also shown positive progress in nAMD and DME, with Phase II data demonstrating sustained visual benefits over two years of follow-up.

TONACEA 03: Building a Moat with Dual Platforms

Over the past five years, AAV gene therapy has experienced a full hype cycle: from feverish capital influx to events like Pfizer's termination of its hemophilia gene therapy, an industry-wide bubble-bursting process.

However, the inherent advantages of ocular tissues – small size, controllable vector doses, relatively closed environment, and suitability for local administration – make ophthalmology the most likely field for AAV gene therapy to first achieve success.

FT-002 and FT-003 represent two typical pathways in ophthalmic gene therapy. The former targets a rare genetic disease with "no approved therapy," while the latter aims at a large indication with "existing therapies but poor patient compliance." Advancing both pipelines to Phase II and presenting positive data at an international conference marks a shift for Chinese biotechs from following to running alongside global leaders in the ophthalmic gene therapy race.

Behind this progress are Frontera's two in-house platform technologies.

On the R&D front, the EXACTE™ platform is an integrated rAAV R&D system focused on three core areas: capsid engineering, payload engineering, and candidate drug screening. Through in vitro and in vivo screening systems, the platform precisely optimizes capsid tropism, enhances compatibility with Bac/Sf9 production systems, and effectively improves vector titers and production quality.

The AAVANCE™ manufacturing platform, based on the Bac/Sf9 cell culture system, achieves a capsid empty rate of less than 1%. Empty capsids are a critical process impurity in rAAV manufacturing, adding inert capsid load and potentially increasing immunogenicity risk. An empty rate below 1% directly translates into a better potential clinical safety profile. This platform is compliant with current GMP standards and represents one of the few domestic capabilities for such industrial-scale production.

These two technology platforms form a differentiated R&D and manufacturing closed loop, supporting Frontera's fully integrated, in-house development of eight pipeline assets, and laying the foundation for its global rights holding and internationalization strategy.

Frontera Therapeutics Pipeline

Phase III clinical trials for FT-003 in nAMD are planned to initiate in China in Q3 2026. The pace is accelerating to bring this intravitreal gene therapy candidate for nAMD toward pivotal clinical development.

For millions of patients "waiting for the light," Frontera is offering not just progress on a drug candidate, but tangible hope for sustained benefit from a single injection.

In a vast market still dominated by "frequent injection" regimens, whoever can first commercialize a long-acting solution has the potential to define the next decade's standard of care. Based on the data and pipeline, Frontera is already on that path.
